The Breast Radiotherapy Service in the Department of Radiation Oncology at MSKCC focuses on treatingall subtypes of invasive and in situ breast carcinomas in the primary context as well as when these tumors recur in local, regional and metastatic contexts. The service line is among the busiest in the department at the level of patient visits and treatments delivered. The service has developed a wide-ranging research portfolio of clinical, translational, and health services studies that examine the role of radiation in the context of all breast cancer presentations. The service also has constructed clinical guidelines on the indications for radiation and the optimal techniques, across the spectrum of disease risks/presentations. The Breast Tumor Disease Site Clinician-Investigator will provide clinical services for breast cancer patients and engage in research activity advancing the research mission ofthe service, with performance site primarily in theManhattan location but possibly in one of the regional sites. Dr. Singer’s Sarcoma Research Lab is seeking an experienced Scientist to join their NIH funded laboratory dedicated to gene target identification and drug discovery in soft tissue sarcoma through a genome-wide genetic and functional analysis. The overall goal of the lab is to perform a comprehensive molecular genetic and functional analysis of soft tissue sarcoma, so as to elucidate the mutational programs and pathways involved in sarcomagenesis and to identify novel therapeutic targets. Sarcoma tissue samples and cell lines are subjected to a multiplatform genome-wide characterization of expression of protein-coding genes and microRNAs, DNA copy number changes, activating mutations, gene rearrangements and methylation profiling. These data are then used to identify both genetically distinct subtypes of sarcoma and molecular signatures associated with tumor morphology, grade, recurrence, and survival. To identify potential therapeutic targets, our lab screens the genes and microRNAs in these signatures for involvement in proliferation, differentiation, and survival of sarcoma cell lines. Potential targets are validated by functional assays in additional cell lines and in xenograft models. Results will be translated into new therapeutics for soft tissue sarcoma.
