Endothelial cells contain 20-fold more acid sphingomyelinase (ASMase) than any other cell in the body. ASMase hydrolyzes sphingomyelin, which is preferentially concentrated in the outer leaflet of the plasma membrane of all mammalian cells, to generate the second messenger ceramide.
